After uploading a sketch to my Arduino board using an USB cable to my laptop, I can see my Pulse Sensor board got powered up as the red LED got turned on.
However, once I disconnect my USB cable from my laptop and connect to an external portable battery, this module is off as the red LED doesn't light up.
Is there any setting I need to use with external battery for this module with PSO2 sketch?

The problem is the power up issue with the I2C lines. Push the reset line on your microcontroller and you should see the sensor power right up.
